The maize (Zea mays) was modified for resistance to lepidopteran insect pests and tolerance to glufosinate-ammonium herbicides. For resistance to lepidopteran pests, the maize expresses the synthetic Bacillus thuringiensis Cry1Da protein (Cry1Da_7), which binds to specific receptors in the insect midgut and forms pores in epithelial cell membranes, resulting in disruption of gut function and eventual death of susceptible insects. The expressed Cry1Da protein provides protection against economically important lepidopteran pests, including fall armyworm (Spodoptera frugiperda) and sugarcane borer (Diatraea saccharalis). For tolerance to glufosinate-ammonium herbicides, the LM maize expresses phosphinothricin N-acetyltransferase (PAT) from Streptomyces hygroscopicus. The PAT enzyme detoxifies phosphinothricin, the active ingredient of glufosinate herbicides, through acetylation, thereby preventing inhibition of glutamine synthetase and allowing the plant to survive herbicide application. The PAT protein was also used as a selectable marker during the development of the transformation event.

Information on the inserted DNA sequence
EH913 corn was developed through Agrobacterium tumefaciens-mediated transformation of the maize Hi-II line using plasmid pEH001, which contains the backbone of plasmid pTF101.1. The inserted transfer DNA (T-DNA) comprises two expression cassettes encoding the insecticidal Cry1Da protein and phosphinothricin N-acetyltransferase (pat).
(1) Expression of the cry1Da_7 coding sequence, which is codon-optimized from from Bacillus thuringiensis strain 1132C, is directed by the ubiquitin gene promoter derived from Zea mays and nopaline synthase transcription terminal polyadenylation site from Agrobacterium tumefaciens. The expression cassette also contains the first intron from Zea mays. High levels of transcription are expected due to the strong constitutive promoter.
(2) Expression of the phosphinothricin N-acetyltransferase (pat) coding sequence from Streptomyces hygroscopicus is directed by a double enhanced Cauliflower Mosaic Virus (CaMV) 35S promoter and terminated by the soybean (Glycine max) vegetative storage protein (TVSP) terminator. The bar gene encodes phosphinothricin N-acetyltransferase (pat), which detoxifies phosphinothricin through acetylation and confers tolerance to glufosinate-ammonium herbicides. The PAT protein was also used as a selectable marker during transformation.
Note
- The cry1Da_7 coding sequence is codon-optimized for expression in maize and encodes a truncated Cry1Da protein (70 kDa) corresponding to the N-terminal 625 amino acids of the native Cry1Da protoxin.
- Whole genome sequence analysis (both Illumina and PacBio), PCR, and bioinformatics showed the integration of a single T-DNA insertion locus containing both expression cassettes in the expected arrangement. An additional cytosine residue in the ubiquitin intron sequence was found but had no impact on any coding sequences. No plasmid backbone sequences were detected outside the inserted T-DNA region.
- Six putative open reading frames spanning the insertion junctions were identified through bioinformatic analyses but are highly unlikely to be transcribed.
- Segregation ratios using chi-square analysis showed Mendelian inheritance across multiple generations, which was demonstrated through inheritance testing across three generations in the L3 inbred line.
- The size of the genetic elements present in the LMO is not publicly available.
- The phosphinothricin N-acetyltransferase (pat) gene is 21 kDa and is also known as the bialaphos resistance (bar).
